Knee injuries to Bridgette Holy and Morgan Dingwall mean the pair are likely to miss the entire season, while Ellie Cuttriss is expecting a child.
But coach Linda Rasmussen is looking for positives in the situation.
“There is opportunities for others to stand up and play well in our A grade side,” she said.
“No team in the competition is going to be able to replace their three best players with ease but we are going to give it our best shot.
“I'm confident the girls we have in our squad will be competitive on a weekly basis.”
Despite the challenges facing the Tigerettes, the absences should allow some of the club's best junior players to thrive.
With several under 15 premiership players moving up an age group, Rasmussen said fans could expect to see them in the senior grades.
“Our under 17s are going to be a really strong side this year,” she said.
“There's some really talented girls in that group and it would be great if they're able to play some senior netball.
“Having those girls play against the best players in the GV is only going to help with their development.
“I'm really looking forward to seeing what they add to our side.”
Rochester are scheduled to play Mansfield at home in round one on April 4.
